toxic buffy coat in tissue culture

We are trying to grow CMV from buffy coat cells from patients
(mostly renal transplant patients) in Hel-cell tissue culture.
After about 2 weeks the Hel-cells die of toxicity. We use dextran
to isolate the buffy coat cells from heparin treated blood
samples. What could be the cause and how do we remedy this toxic
fenomenon? 
To investigate some possible causes of this problem we used
different dextran batches and we tried blood samples from healthy
donors. All of these produced the toxic effect. Other sample 
types
(like saliva) did not have toxic effects.
Suggestions, please.
