I'm just starting up with Staden, and I have a quick question. All our
clones are routinely sequenced in both directions. Do I need to run pregap
on them in 2 batches, specifying different offsets for the different
sequencing primers? I'm a little unclear as to how the direction is
determined in pregap as well. Should I specify one offset as being negative
(since it is the other side of the cloning site)?
