Your question is far too general to really answer.  Yes, serotonin
reuptake is an important mechanism to re-establish presynaptic
serotonin stores after release.  The presynaptic terminal also
synthesizes serotonin and that also refills the system. 

The real question is why you would suggest alterning the natural
process.  If things are working normally, then there is no need to
change anything.  If things are going wrong, then you have to look at
just what the problem is before trying to fix it.  The problem could
be failure to synthesize it, problem to release it, problem to respond
to it postsynaptically, problem to recycle it. Usually, a "deficiency"
in serotonin response is treated with a reuptake inhibitor, not a
reuptake stimulator. By inhibiting reuptake, you make the serotonin
that is released stay longer in the synaptic gap and therefore exert
more of an effect.
