Hi Mike!
50 microns is not bad for a piece of brain whit the size of 5-6
sq.inches.
I will recomend you to play first whit the temperature.Set it low
about -25,-30 C,It depends also what type of cryostat you are using
.For big sectioning is also good to use vibrating microtom.
