I don't know about single layers, but if you stain with propidium iodide
(PI), the result appears very similar to a Nissl stain. Experienced
anatomists can perceive the layers in such stains. PI has broad
absorption and emission spectra, so you have to stimulate it separate from
the other channels, but it works.
