Usually E. coli is being sent in soft agar in a stack. Just make rich
media with less agar (I think 0.5%) and pour it into a sterile cryogenic
tube. Pick a colony from a plate with a bacteriological needle and stick
into the media. Let it grow o.n. and sent it.
BTW The E. coli stock center sent their stuff on filter disks - the main
advantage it can be sent in a regular envelope no need for package.
