> Does anyone have a good culture medium for growing Volvox sp.?
As funny as it sounds, when I was a kid I used to get great Volvox
growth in the 'fishmeal' medium, which consists of 0.25 gm fish meal
autoclaved in 500 ml. of distilled water. After autoclaving, add sterile
solution of Ferric chloride (1ml of 1.0% solution), sterile filter and
use.
As it turns out, this medium is also mentioned online at
http://gwis2.circ.gwu.edu/~biodl/technique/Fishmeal.html
(though I would still autoclave the stuff, rather than heat to 80C).
Volvox media is available commercialy, item #L261A at $3.95 a pint (!)
from http://www.southernscientific.com/media_handling_aids.asp
As well as actual Volvox cultures, from
http://www.berkshirebio.com/store.html
Finally, if you wish to go completely synthetic, use:
http://www.bio.utexas.edu/research/utex/media/volvox.html
(Provasoli & Pintner 1959).
Not suitable for xenic cultures. For additional modifications of volvox
medium, see HEPES-volvox, MES-volvox, P49, volvox-dextrose and YT20.
