I am hoping someone out there can help me. I am trying to electroporate
a very large P1 phage plasmid that is approximately 80kb in size into
E.coli competent cells. I have only 0.1 and 0.4 cuvettes at my disposal,
no 0.2 cuvettes but I don't think that is the problem. Right now I am
trying different amounts of DNA and cells, and different voltages, but
with no success so far. Does anyone have any knowlege that can shed some
light on this problem? I would be very grateful.
