You can measure spore-functions directly after rehydration as they
respond then to normal membrane integrity measurements. My method of
choice would be flow cytometry (FCM). The Army has develloped a number
of methods to quickly germinate spores. Once the rehydration has occured
dead spores are recognised by impaired membran integrity. Unfortunatelly
even of all the spores that have undergone phase change and are intact
not all will elongate or polarize and replicate. Also there will be a
remaining fraction of spores that do not even undergo the phase change
in the first place. They can potentially also be quantified and tested
for extracellular enzyme functions using FCM and fluorescent probes.
