A range of organisms should be used for this type of evaluation. As a =
recommendation I would suggest using those stated in the US Pharmacopeia =
for challenge testing ie. S. aureus, E.coli, Ps. aeruginosa, C. albicans =
and Aspergillus. The best media to use would be those recommended for =
antibiotic sensitivity testing eg. Mueller Hinton, Nusens or Sensitivity =
Test Agar, as they do not contain antagonists.
The disc diffusion method may not be the best for this application as =
the oils will not diffuse well. It may be better to use a standard =
challenge test or preservative efficacy test. These involve inoculation =
of a known number of organisms into a solution containing the =
anti-microbial. The mixture is then sampled at intervals and a decrease =
in numbers should be observed.
