I am trying to do insertion mutagenesis by triparental mating, and is
wondering which frequency of single crossovers might be expected.
Recipient strain: Rhizobium meliloti,
Donorstrain: DH5alpha with 8 kb plasmid (370 bp "almost homologous"
sequence, reportergene, Amp and Kan resistence genes ++) to be inserted
into the R. meliloti genome
Helperstrain: DH5alpha pRK2013
And, what frequency of spontanious resistense to Kan and Amp should I
expect?
