Can anyone offer any advice regarding isolation of type II alveolar
epithelial cells from the rat lung?
I have been attempting to follow the method of Dobbs and others (1986) but
have experienced major difficulties.
I have tried using Elastase from both Sigma and Worthington and delivery
of the enzyme to the lung via the trachea or after chopping the whole lung
but can only liberate a very low number of cells and a low percentage of
type II cells (30%) in this inital crude cell suspension.
Can anyone describe in detail the conditions required for successful
panning on IgG coated plates?
Any suggestions would be much appreciated.
