How to circumvent DNA restriction in a bacterial host?

	Dear Jim:

	From my experience, you have two options. You could try 
mating your construct into your recipient via conjugation from an r-m- 
strain of E. coli. This would tell you whether you have a restriction 
problem or a transformation problem. A second approach entails 
isolating a restriction minus strain of your recipient. We did this with 
Legionella and it is relatively easy. If you would like details, please
