Growth of Propionibacterium acnes

If you want to isolate (obtain high colony counts) of 
Propionibacterium acnes on human skin, you should use 5% (horse) 
blood in Columbia agar. You will than get many colonies, but small 
colonies after 48h anaerobic incubation.

If by any chance you would like to have high yields (mass 
production) of the organism, you should use liquid medium, 
start with high inoculum and increase oxygen concentration 
slowly. P.acnes is VERY oxygen tolerant.
