Subject: smearing on pulse field gel
Hi netters,
I am working on genomic mapping of Pseudomanas by pulse field gel
electrophoresis. One particular strain of a species I'm working with
produces a smear when digested within agarose plugs whereas other
strains of the same species can be cut successfully by the same rare
cutting enzymes such as SwaI or CeuI. However, they all can be cut by
SpeI. Does anyone have any suggestions? Any help at all will be greatly
appreciated.
