Inoculate 2 tubes of Hugh Leifson media. Overlay one with sterile mineral 
oil (or strictly speaking you should use sterile petrolatum to insure good 
anaerobic conditions). Incubate for 48 hrs or in some cases it may take up 
to 14 days for reactions to occur. If both tubes turn acidic the organism is 
considered a fermenter, if only the tube without oil turns acidic the 
organism is considered an oxidizer and if no acidity is detected in either 
tube, the organism is considered a nonutilizer of the sugar. Hope this helps.
