How do you know the plasmid is there to start with?
That really isn't a rhetorical question. But basically you need to grow
cultures up under curing conditions, such as with acridine orange or
ethidium bromide, and then test lots of individuals colonies for the
plasmid. However it is that you know the plasmid is present is how you
need to test for it. This could be resistance or sensitivity to some
phage, ability to mate with
some other plasmid, or just run lots of gels and look for plasmid DNA.
