In response to the query by Tommy on the composition of Pseudomonas biofilms,
there is quite a diverse literature. With regards to the structure of
biofilms, recent articles by Wolfaardt et al. Appl. Environ. Microbiol. 60:
434-446, 1994; and Lawrence et al., J. Bacteriol. 173: 6558 -6567, 1991 employ
confocal scanning laser microscopy and computer imaging to construct three
dimensional images of living, fully hydrated biofilms. Previously, biofilm
architecture had been largely described by electron microscopy. When one
considers that the extracellular capsule material (which is the dominate
feature of biofilms) is ca. 90 - 95% water; many dehydration artifacts arose
during preparation for electron microscopy.
