I don't think a single thaw-refreeze cycle will have a major detrimental 
effect on culture survival.  We have some glycerol-frozen preps which 
have gone through several (three or four) of these cycles and have 
maintained their viability.  

However, if you're dealing with the last vial of an irreplaceable strain, 
you might want to think twice about doing this... :^)
