THE CARBOHYDRATE RESEARCH PROGRAM AT UTRECHT UNIVERSITY
The integral research program of the Carbohydrate Section of the Department
of Bio-Organic Chemistry, Bijvoet Center for Biomolecular Research at
Utrecht University is focused on structure analysis (I), synthesis, (II)
and interaction studies (III): (I) The study of polysaccharides and
glycoprotein glycans in terms of their primary structure and
three-dimensional structure in solution, and the study of intact
glycoproteins in solution in terms of influence of the carbohydrate chains
on the protein conformation and vice versa (primary structure analysis
glycoproteins, e.g. recombinant/transgenic glycoproteins, glycohormones,
human amniotic fluid glycoproteins; primary structure analysis
polysaccharides, e.g. exopolysaccharides from lactic acid bacteria, plant
cell wall polysaccharides; 3D analysis (NMR and molecular modelling), e.g.
human chorionic gonadotropin, exopolysaccharides of lactic acid bacteria).
(II) The organic / enzymatic synthesis of oligosaccharides, which form
structural elements of polysaccharides and glycoprotein glycans
(oligosaccharide-conjugate vaccines against pathogenic bacteria,
oligosaccharide-conjugate diagnostics for schistosomiasis). (III) The study
of interactions of carbohydrate chains and complementary molecules on the
molecular level (surface plasmon resonance, trNOE NMR spectroscopy,
molecular modelling; milk oliogosaccharides; marine sponge
oligosaccharides, alpha/beta chorionic gonadotropins; lectins).
The research group comprises at the moment besides the two group leaders,
Prof.dr. J.F.G. Vliegenhart and Prof.dr. J.P. Kamerling, three staff
members, six post-docs, and seven Ph.D. students.
The Bijvoet Center is well equipped for studies in structural glycobiology.
A great variety of separation techniques is available, e.g. convential
chromatography, FPLC, HPLC, HPAEC, capillary electrophoresis, GLC.
Furthermore, advanced NMR (up to 750 MHz), advanced MS (including the
modern ionization techniques), and biomolecular interaction analysis
(Biacore biosensor technology) instruments are available. All microchemical
and enzymatic techniques in glycoconjugate analysis are applied.
POST-DOC VACANCY IN SURFACE PLASMON RESONANCE (TWO YEARS)
Surface Plasmon Resonance (SPR) is one of the technologies used in our
group to study carbohydrate-protein and carbohydrate-carbohydrate
interactions. Typical recent examples from our research comprise
lectin-carbohydrate interactions (e.g. ConA, SNA, MAA), monoclonal
antibody-carbohydrate (as neoglycoconjugate) interactions (e.g. from the
synthetic carbohydrate-based Streptococcus pneumoniae vaccine project),
carbohydrate-carbohydrate (as neoglycoconjugates) interactions (e.g. from
the marine sponge Microciona prolifera project).
The present responsible post-doc, who has run the Biacore instrumentation
during the last three years and who carried out all the mentioned studies
in joint collaboration with other post-docs and PhDs, involved in the
preparation of the starting materials, in our group, has accepted a
position in his homeland. This means that we can offer this, from a
scientific point of view, exciting job to a new post-doc, who is interested
to explore this highly promising method in more detail by studying
interaction phenomena from the structural glycobiological field.
The salary of the qualified person will be according to Dutch University
regulations.
