For MS analysis (at least using MALDI-TOF), there is no need to desialylate
the glycoprotein, though sialic acid containing glycopeptide may have some
problem for detection. I have successfully determined bi-antennary
complex glycopeptide with 0, 1, and 2 sialic acids based on Con A binding (
10 mM alpha-methylglucoside elution), sialidase and galactosidase digestions,
and MALDI-TOF analysis.
