Recombinant glycoproteins expression in insect cell....

Dr. Chang

Your question is a very important one, which has been addressed by the
superb studies of Dr. Francis Castellino and colleagues at Notre Dame
University. They report that twenty hours post-infection with
baculovirus, Sf9 cells produce recombinant glycoprotein bearing only
simple truncated N-linked structures with terminal mannoses. However, as
time after infection progresses, the complexity of the N-linked glycans
increases, until at 60 hours post infection there are typical
tetra-antenary oligosaccharides with terminal sialic acids.

An excellent review and example of these studies was published by F.J.
Castellino, et al. in Methods of Enzymology 223, 168-185 (1993). See
especially the structures on pp. 180-181 and the Tables on pp. 182-184.

Being a bit removed from this area, it has been my impression that Dr.
Castellino's findings have not been as widely appreciated as they might be
and your question is an excellent opportunity to discuss the work on the
net. Can anyone else comment?
