We are using the Oxford Glycosystems Glycoprep 1000 automated
hydrazinolysis instrument for release of oligosaccarides. Our recoveries
off of the instrument are between 20 and 40% based on monosaccharide
analysis before and after release. We also obtain similar recovery numbers
by Fluorophore Assisted Carbohydrate Electrophoresis compared to the
theoretical value. Has anyone else looked at recovery of oligosaccharides
from the Oxford Instrument? If so, how do your numbers compare and how do
you determine recovery. Does anyone have any suggestions for an internal
standard for this system.
