I have a couple of questions about analyzing autoradiograms of 35-S labeled proteins seperated by 2-Dimensional PAGE. What I would like to do is to quantitate the amount of label incoporated in various spots and compare these levels between gels. My questions are: 1) what is the best (and preferably cheapest) software to do such analysis (I already have a 1200 dpi flat bed scanner), 2) I've been told that I must pre-flash the film to get a linear response.
