Has anyone ever observed sequences which start out perfectly well (about
twenty bases) then give what looks like tens of Ts, then nothing. This
is _NOT_ polyT, but looks like it could be due to contamination of
something on our platform as hand-made sequences on the same templates
work fine.
We've observed this problem on sequences prepared on an Amersham Vistra
robot and a Tecan Genesis, both using MilliQ water. We use BigDye
version 3 sequencing kits and Millipore MAHV Sephadex columns for
purification. We've observed the same phenomenon on ABI377 and 3100
machines.
