I am currently in the process of evaluating the pro's and con's of a
megabase vs. a 3100 platform for DNA sequencing.
In particular, I would be interested in answers to the following questions :
1. Comparative read lengths : I am led to believe that the megabase with
its proprietery polymer yields longer reads than AB platforms. Is that
still true for a range of template types, differing in quality and/or
quality ?? I would be particularly interested to know if anyone has
information regarding version 3.0 big dyes and different types of
templates, varying in quality, quantity and base composition on the 3100
vis-a-vis the same material on a megabase sequenced with version 2.0
thermosequenase dye terminators. Comments on either platform in this regard
would be very helpfull. Also does anybody know the maximal read length
attainable from an ideal template such as ( 200ng of ) pGEM with either
system ??
2. Reliability
3. Number of runs per day with either platform and the number of
samplesanalysed per run.
4. The range of template conc. conducive to yielding good data : I know
from using 377's and 3700's that 10 ng - 1ug of template is optimal for
templates ranging in size from 100bp - > 10kb, including BAC's,
respectively. For the 3100 in particular does anybody know if these figures
hold true for version 3.0 big dyes, I suspect they do ( ?? ).
