Alison,
This possibly may be one of two things,
1) the laser isnt returning to the start position before the run is
started, restarting the computer and rebooting the program should help
this (the latest version of the software should deal with this)
or
2)If your losing lanes off both sides of the gel at the same time its
probaly due to the plate cleaning regime not being strigent enough
