I have been given the uneviable task of having to sequence a cosmid. The cosmid is pWE15 from stratagen and has a 3kb insert. having never sequenced any larger
than a big plasmid (10-11kb including insert). I was wondering if anyone has any special hints or stratagies for sequencing a cosmid. Do I need to add Mg or
change cycling parameters, longer/shorter annealing times.
Any help is greatly appreciated
