Hi Barbara: You mean you haven't thrown out all your 377's and switched
over to capillaries? Just kidding, but how is your capillary working?.
We have been using the 96 lane 377's since they came out (when was that,
two summers ago already?). We lose about 50 bp in phred Q>20 bases, but
the increase in throughput and ease in loading a 96 well format more
than compenstates up for that.
We are using Gel Company plates, PEB spacers and combs, and Kloen
loaders. We add matrix stds to lane 1 48 and 98 so we can find the
right, middle and left sides of the gels. The new sofware does a much
better tracking than the old. The only real problem is where you have a
bunch of failures or too much signal, then it gets hard to call your
lanes. Capillaries make that a moot problem.
We haven't had the problems other people seem to have had or we decided
that we could live with them. Guess we were lucky (where's some
wood????).
