Filter combs & loading protocol

Hello,

I have been using 64-lane paper combs from the Gel Company for GeneScan
analysis. I use a modified water-loading protocol. I load the samples
with 0.5x TBE and then add more TBE to equal 1X for the rest of the run.
This has given me much better lane separation than straight water-loading.
I tried 96-lane combs with water loading and ficoll loading with very poor
results, but I had other problems too. Next time I will try my 0.5x TBE
loading.
