I'd like to get some tips from anyone that is successfully sequencing
plasmid inserts with Pharmacia's ALF. In particular which sequencing
enzyme
works well, which protocols to use for labelled primers (which primers are
best) or internal labelling. I've tried Pharmacia's Autoread kit without
much success lately, and more recently Amersham's Thermosequenase has
worked quite well for fluorescin labelled primers (here I'd like an
internal labelling protocol if possible). All I want is 600+bp of good
sequence each time. It's not too much to ask, is it?
