I too sometimes use half volume reactions with our ABI 373Stretch
machine. A
couple of hints though: If the template quality is variable (as ours
are) the
results can be dissapointing. The read length is usually shorter as
well.
Try using the halfTERM reagent from GENPAK, it's really cheap, it allows
you
to use half of the enzyme amount and it greatly improves the read length.
We
usually get over 800 bases when using the halfTERM reagent.
