Is there anyone who used yeast one hybrid kit ("matchmaker one hybrid library construction and screening kit") from clontech. I tried to prepare SD -leu DO medium supplied with the kit (cat # 2684). There is mentioned 0.69g DO supplement + appropriate min. SD base/1L on the vial labell. To make solid media what should I do? Should I dissolve this powdered DO supplement directly to water alongwith 2% agar or I have to add SD base media seperately? I added 0.15 g D/O supplement provided with the kit and 0.3g agar in 15 ml medium but the media did not solidify.
