Hi everyone
the GenBank sequence of pRS315 predicts two XhoI sites, one in the
polylinker and one between LEU2 and f1. However, the Sikorski and
Heiter paper says that the XhoI site is good for cloning into the
polylinker. Any thoughts? (the direct experiment says that XhoI only
linearizes the plasmid).
