Hi

I'm trying three hybrid system with pBridge plasmid(Clontech).
The plasmid have Met 25 promoter which is activated in the absence of
methionine.

I constructed some plasmids with pBridge and transformed into AH109
strain.
But the cells could not grow in the absence of methionine.
I used SD medium (-Trp, -Met).

I'm not specialist for yeast research so I need more information.
Can S. cerevisiae grow in the absence of methionine?
Do I need to add anything as substitute for methionine?

If you have some experience using pBridge,
Please tell me your case.
