DNA isolation and PFGE

Can anyone recomend a protocol for isolating the 3 pombe 
chromosomes within an agarose plug (for use in pulsed-field gel 
electrophoresis)  I'm using the method in Nurses fission yeast hand 
book at the moment but would like to play around with a few more 
methods.
