I am currently trying to do a one-hybrid screen using a cDNA library cloned
into a pB42AD library. In order to have expression of this library, I must
use a galactose-responsive yeast strain. However, I must also integrate
two reporter constructs that are selectable for His3 and Ura3. The only
strains I know of are either gal-nonresponsive or are completely deleted
for his3 and ura3 (a case that will not allow homologous
recombination/integration of my reporter constructs). Does anyone know
where and how I can get a yeast strain that is wild-type for galactose
synthesis and that has a partial deletion for his3, ura3, and trp1?
