Hi, everyone.
I am looking forward to finding some pcr primers for gene
disruption of saccahromyces cerevisiae, for example, hsp30,
60, 90, 104, 70, NTH 1, tps1...related on yeast stress
responses.
till now I have constructed these primers using the primer
program in SGD, but the results were not good, multiple
bands or no band.....though almost all experiments had been
done based on the pcr troubleshooting guide.
What can i do solve for these problems? if you have some
these primer sequences on the gene related on stress
responses, please tell me the primer sequences ..
My strain F38-1 has an outstanding intrinsic thermotolerance
and shows various intriguing events like as hsp inductions
and membrane fluidities .......For revealing this events, I
have to construct the mutants so I need your help.
