Dear colleagues,
We have just performed a yeast two-hybrid screen. Some of the
clones we pulled would make sense, however, there is a termination codon
between the coding sequence of the activation domain and the long open
reading frame of the (putative) interacting protein. To produce the fusion
protein, ribosomes would have to skip over this termination codon. Is this
possible? Someone from Clontech told us that this can happen, but I would
like to get more feedback before we invest time analysing these clones.
