I am trying to detect pairwise 2-hybrid interactions by mating.
The binding domain hybrids are in alpha cells and the activation domain
hybrids are in a cells, after mating diploids are screened for 2-hybrid
signal. Does anyone know if the ploidy of a cell might affect expression
of the hybrid proteins or the two-hybrid interaction? The hybrid proteins
are constitutively expressed from CEN plasmids. Thanks for your help!
