YPD medium lacking uracil

Allow me to re-phrase. A strain that cannot salvage pyrimidines
(5-fluorouracil resistant, e.g. uracil phosphoribosyl transferase deficient
[fur1]) cannot use exogenous pyrimidines regardless of the type of medium
(YEPD or synthetic medium). It follows that a fur1 strain is completely
dependent for growth upon de novo pyrimidine synthesis. A fur1 strain that
contains a chromosomal ura3 mutation *and* URA3 on a plasmid is absolutely
dependent for growth on the URA3 plasmid -- it dies if it loses the
plasmid. This has nothing to do with 5FOA.
