We are analyzing the determinants of transmembrane (TM) Protein
Insertion Orientation (PIO) in yeast. Initial results (JBC , 1996, 271
24625-33) established the validity of the charge difference rule. We
analyzing charge-independent determinants and the attenuation of the charged
signal by distance. A refined rule for topology prediction will be
applied to the yeast proteome database.
Mutants have been isolated in which PIO is relaxed. We
cloning will identify the components of the receptor responsible for
PIO, a primary event in protein translocation at the ER.
