A restriction enzyme method with gel recovery has been published by
Wiesner et al., Gene 98:277, 1991; basically bulk DNA is cut with enzymes
which don't cut mtDNA, and the latter is recovered either in linear or
circular form. It is pretty easy but requires alot of genomic (i.e.
nuclear + mtDNA), ~20 ug.
