carry-over of medium colour

pichia supernatants are replete with colored compounds, small (metals) 
medium (flavins) and large (proteins). in my experience they can be 
removed by a chromatography step at the beginning, say anion or cation 
exchange (others will also work). recloning should not be necessary, 
because there are very few proteins in the supe, and tagging is 
redundant.
