Direct PCR of yeast clones picked from agar plate?

I use a modification of the technique in BioTechniques 19:744-748 (1995). 
I swirl a small amount of a yeast colony in 2 ul of 3 mg/ml Zymolase and
incubate at 30 deg. C for at least 15 min, then add this to my PCR
reaction.  I have had excellent results for both S. cerevisiae and P.
pastoris.
