Hi everyone,
We have had quite a few letters regarding the protocol members in my lab
now have working for Northerns. It is in the manual "Methods in Yeast
Genetics" A Cold Spring Harbour Laboratory Course Manual. 1994 edition
by C. Kaiser, S. Michaelis and A. Mitchell. It is quite a long protocol
but I am assured it is worth the extra effort. With regards to the
herring sperm DNA, it was too big (>300bp) so get some that you can shear
to below this (we now use some grungy looking calf thymus DNA and it
seems to work). I have yet to personally get this going, but I am
confident having observed the nice results of others in my lab.
