Thank-you to everyone regarding the brilliant advice on Northerns. It
would seem we now have a method which is working well (Cold Spring
Harbour method) and we believe the problems were partly due to the size
of herring sperm DNA we used to prevent non-specific binding and also our
washing protocol (needed to be VERY stringent to get rid of the
non-specific binding to the rRNA). I wish everyone well with their
research and will keep my eye out on the net for whenever I can return the
favour!
