Hi yeast netters,
I've got an unknown Saccharomyces cerevisiae gene of interest fused to the
reporter gene lacZ. Unfortunately the plasmid had been integrated into the
genome making it difficult for me to pull it out. I thought of using a lacZ
primer to sequence whatever is fused to it. If anyone has had success with
yeast genomic sequencing could you please give me some advice on making
clean genomic DNA preps, primer length etc. I am using PCR for sequencing.
