Hi, any amplifiable vector cognoscenti out there?
I am trying to put together a sequence map for the amplifiable
YAC arm vector pCGS966 (Smith et al, PNAS 87: 8242-8246),
but the source references don't seem to be able to clear up
one specific point, which is where exactly did the HSVTK
portion come from? A plasmid called pAYE56 is cited, but
published details of construction of pAYE56 are scanty and don't
cover the TK-bearing progenitor pAYE81.
Also, does anyone have sequence for the "missing" Trp promoter
sequence that was added back into pCGS966? I can't find
anything in the GenEmbl database.
