Hi,
I am new in the yeast field and have just a silly problem. I have
transformed a leu2-3,112, his3-11,15 strain with a replicating plasmid
containing a HIS-reporter gene and a LEU-marker. I have got only very few
colonies (4), compared with a control plasmid without the HIS-gene which
gave several hundred colonies. I have transformed with the same amount of
DNA. Is the HIS-plasmid not stable in this strain, or could it be
integrated in the HIS or LEU locus ?
